Fig. 8.   Effect of protein phosphatase inhibitor, okadaic acid (OA), on secretin-induced expansion of area of luminal space of IBDU. A: OA alone (bullet ; 500 nM; n = 4) failed to induce significant changes in baseline area of luminal space during 30-min (time 15-45 min) exposure, and showed no differences compared with IBDU perfused with KRB only (black-square; n = 7). B: secretin exposure for 10 min induced a progressive expansion of luminal space up to a maximum of 65 ± 18% of initial values (35 min); thereafter, luminal space fell to 23.9 ± 6.9% (n = 6). In contrast, when IBDUs were perfused with OA (between 10 and 50 min), initial exposure to secretin (between 10 and 20 min) induced a progressive expansion of luminal space up to 97.9 ± 19.1% of initial area (n = 8; P < 0.02 vs. secretin only), indicating persistence of secretin stimulatory effect. black-square, Secretin alone; bullet , secretin + OA.
